Position-effect variegation in Drosophila: A genetic and molecular approach of epigenetic silencing

Epigenetics, the heritable changes in gene function occurring without a change in the DNA sequence is a major new field in life sciences. Here again, Drosophila has led the way with work starting almost a century ago with position-effect variegation, a phenomenon of mosaic expression of genes relocated near heterochromatin by a chromosomal rearrangement.

Genetic modifiers of position-effect variegation have identified essential components of heterochromatin-induced epigenetic silencing as predicted by Reuter and Spierer (1992). We have molecularly identified a number of modifiers of position-effect variegation. The genes we have identified encodes heterochromatin proteins ands modifiers of histones (methylation, acetylation, and ubiquitination). We have devoted a lot of work on one of them SU(VAR)3-7, and have revealed that it participates with other partners in the building of heterochromatin resulting in the gene silencing of position-effect variegation. We have characterized its function by analysing the phenotypes of Su(var)3-7 mutation and over-expression in soma, dissected the different domains of the protein and described its implication in heterochromatin formation. We pursue or study of SU(VAR)3-7 function with different partners in different pathways. There are four main lines of research:

Transposable elements as targets of SU(VAR)3-7 in heterochromatin

We have made recently two important discoveries. First, SU(VAR)3-7 interacts with dosage compensation, the mechanism up-regulating expression of genes on the unique male X chromosome to make it equivalent to the two X chromosomes in females. This important phenomenon depends on the proper dose of SU(VAR)3-7. Second, we found that SU(VAR)3-7 controls the activity of a number of transposing elements. Here again, the animal needs to control part of its genome whose activity might be detrimental.

SU(VAR)3-7 and its heterochromatic partners

After a comprehensive analysis of SU(VAR)3-7 physiology in the soma, we describe in details the phenotypes of Su(var)3-7 mutations in ovaries, on oocyte differentiation, germ line chromosome integrity and inheritance. We will expand our study to other actors of the chromatin as the H3K9 HMTase dG9a and other Su(var) genes.

The roles of DmSETDB1

We have discovered that the Drosophila methyltransferase SETDB1 is required for chromosome 4 silencing, and we pursue the dissection of this phenomenon. We also analyse the activity of DmSETDB1 on target genes and we identify its partners.

In sum, we have discovered several actors of gene silencing in epigenetic control of gene expression, in the regulation of transposable elements and in chromosome integrity and segregation at mitosis and meiosis. We foster this analysis with a special emphasis on the female gonad. In addition to contributing to the expanding field of epigenetics in biology and human diseases, we expect that our work will bring novel understanding on the so far poorly explored role of epigenetic mechanisms in germline differentiation.
